Volume estimation

Hello!
I'm trying to estimate the volume of lesions of cortical impact injuries in rats via tomographic reconstruction. I surrounded the lesion, creating a ROI, to get the area. For estimating the volume I'm using the MeasureStacks plugin. There I have the problem that I don't have a constant Slice Spacing, the distance between the slices differs. Does anybody know a possibility how I could measure the volume anyway?
Furthermore I wanted to do a 3D-reconstruction of the lesion but don't succeed in rendering only the ROI I marked and - again - I have the problem with the distinguishing Slice Splacing.

I'd really appreciate if somebody could help me!
