Hi Huiming,
Thanks for providing an image, it's very helpful. Your outlines are wide
enough, so don't mind what I said about binary open or close. You just
need to convert your color image to a binary (black & white) image; try
this:
1. Process > Binary > Make Binary
2. Edit > Invert (you may not have to invert but do try it the first time)
3. Analyze > Set Measurements, then select "Area" and "Shape descriptors"
and click OK.
4. Analyze > Analyze Particles, here I'd use Show Outlines, Display
Results, Clear Results, and Summarize.
