cell counting

Hi!
I have been trying to do automatically count cells in digital image (tif) for brain slice but am having trouble. My samples are human and I used NeuN antibody with DAB staining. I would like to measure the dark cell nuclei. I don't have lot of experience imaging. I followed the ITCN movie but this plugin didn't write the cell number in the end. Then I repeated it an fluorescent picture which is working. The ITCN work only with fluorescent pictures? I attached my picture.
