3D counting in a ROI

I need to count and measure particles inside a ROI, in a z-stack. What I actually need to do is to estimate the  number and dimensions of Golgi elements per cell, using z-stacks. So I want to manualy delimitate cell limits and set this as a ROI, and measures the Golgi elements inside these ROIs.
