Calibrate and Measure Freeform Line

Hello all,

I am new to ImageJ and new to this listserv as well.  
I am trying to measure the length of epidermis in a skin section, which is a slight curve with undulations.  I also need to calibrate the software using a 1mm slide micrometer.  

Can anyone point me in the right direction for these procedures?
