Measuring increase in size from video

Hi, I have a video containing roundish subjects and want to measure their increase in size over time. My idea was to take screenshots of the video every X minutes and analyze. Initially, I thought I could use color threshold as a standardized way to detect the shapes (and increase in area) each time, but the contrast difference between my shapes and background is poor and I haven't had any luck. The outlines are clear to the eye though..

Any ideas? If someone would be willing to take a look at my image and give me some advice, I would really appreciate it. I'd prefer to send in a private email though. Thanks so much.
