Correcting optical offset

Hi there,

I am measuring the distance between two red and green gaussian fitted curves. I then repeat on more fields to get an N= aprox 500 and calculate an average (I am using a macro to automate this).

I think that the red and green pixel offest is increasing the standard deviation of the calculated average. I would like to measure the offest (presumably using beads that flouresce in both channels), and then re-run a modified version of the macro that corrects for it.

Are there any macros out there already that do something similar that I might be able to adapt?

Can anyone suggest a good route forward?  I am new to writing macros, and not yet very good at it, so help would be gratefully received.
