NeuroJ-Calibration

I am currently working with primary neuronal cell culture from mouse cerebellum and I'm using NeuroJ (An imageJ plugin) for analysis of neurite outgrowth. But before proceeding with the analysis I need to know what are the necessary steps to do to calibrate my measurements?
