How to remove backround

?I am trying to do some area measurements on lung tissue ( HandE sections). I would like to be able to measure the total area of lung tissue with the Background (empty spaces) subtracted. I can do this very well using color threshold and the brightness sliders. I can select to measure the area of background or the area of tissue and I get very good results!


I would then like to outline and measure area (ROI) of inflammation that may have some "empty space" that needs to be subtracted. This is where I am having a problem. I can't seem to be able to perform the same function on an area as I can on the entire image.


Anybody have any thought?
