set scale as scale bar,but get different results

Dear All, I want to use FFT to transform a EM picture. This picture is not
original. I made screenshots from a paper. But the picture has scale bar. So
i zoom the picture to get different size pictures. I set scale based on the
scale bar on the picture. But i got different distance. Is there someone can
explain this problem? Thanks zhongwu
