The big problem trying to segment the DIC images are the shadow-cast effect. I work a little bit with that problem and found a interesting paper [1] which tries to eliminate this effect. Maybe after remove the shadow-cast effect the segmentation process could be more easy (in my case it was).
I made an implementation of the latter method as a plugin for ImageJ and you can find here [2] if you need it.
In order to use it, you need two extra images, one black (pixel value = 1) and one white (pixel value 255) (see [1] to know more of these images), with the same size of the image that need to process. Also you need to know some of the physical characteristics of the DIC filter attached to your microscope.
