Measuring blood flow through a channel

Hello all,

I have a high-speed time lapse of fluorescent erythrocytes passing through a heart valve that is about 2-3 RBC's wide.  I'm hoping quantify flow rate in the valve through the heart cycle.  I have a vague feeling that you can do this using a kymograph, but I am not positive which approach would be ideal.  Does anyone have a favorite tool for solving problems like this?
