Dear Adrian,

You can use Process > Image Calculator to subtract images. There is also the option to use Process > Subtract Background... what uses a rolling background approach to correct uneven illuminated background, see: https://imagej.nih.gov/ij/docs/guide/146-29.html#toc-Subsection-29.14

I use to substract background in fluorescence images (1 channel) by substracting the mean value setted by measurements. In the case that the background is not uniform i would like to know if it possible to substract the entire image without the ROI from the original image.
