cell counting

Hi

I am new to ImageJ. I am trying to count phytoplankton in an
hematocytometer. The cells, no matter how much I wait, do not sediment.
Further I can clearly notice that as time passes the volume rather quickly
decreases under the slide and this wolud result in wrong counting.

What I did was to take pictures in several planes. I would like manipulate
each of them so as to only observethe cells in focus and then stack all the
different focus plains and then count.

Any suggestions on strings of commands I should execute?
