Isolines - how to make it

Dear experts

I am using IsoPhot plugin to mark the certain pixel values in the image, see
attached.
However, now I need to get an average isoline of the marked areas - in other
word, the plugin/script should the distance between the same points of the
same pixel column and mark the middle point.

Maybe someone can share some experience ?
