Quantifying colocalization and measuring focal adhesion colocalization in puncta

Hello!
What is the best way to measure the changes in focal adhesion colocalization? Do you measure the individual puncta, or whole cell fluorescence?

Additionally, does anyone have any recommendations for quantifying the amount of colocalization in immunofluorescence?

Any help or advice in these issues is appreciated!
