unit for surface value

Hi,

I would like to know at is the unit for the surface value given using the ROI Manager.

I'm trying to get the surface value of synapses. I have an in vivo fluorescent marker for synapses. I take a z-stack of the signal, then project the maximum intensity to have a single image. Then I circled the different dots and extract the different values using the ROI Manager. For the surface I have value between 0.0001 and 0.001. I'm trying to understand what is the unit of this "surface". I read it is in square pixel but my issue is how the surface can be inferior to 1.

Hi Muriel,

On way to know is to check the pixel unit of your image, is it pixel or micron ?

To do so:
- open one of your image in ImageJ/Fiji
- within the menu "Image" go to "Properties..."
A window pops-up with many informations regarding your image, and one is "unit of length".
If it's micron , it's easy, your values are micron^2
If it's pixel, you have to calibrate your image, ie get/calculate the pixel/voxel size.
