Collagen Staining Questions

Hello Image J,

I am learning to use the image J software.  We used Mason's Trichrome Strain
to stain myocites for collagen content.  The cells are red and the collagen
is blue.  I attempted to use the example on the image J website involving
the threshold function.  I have been able to get the collagen highlighted
with the threshold function but I'm not sure where the best threshold value
lies.  Do you have any ideas on how to get the most consistence and complete
% area of collagen?  Is there a better way the the threshold function to
look at collagen?

You should try color cluster segmentation.
