Creating boundary on merged cells

Hi All,

Thank you for involving me in the imagej forum.
I have a query, how could one create boundary/segmentation based on cell membrane ? I am attaching here an image on which I would like to create boundary.
Any help will be much appreciated.

It looks like you stained for f-actin in green which does not mark the cell boundaries.  I would label cell membranes or proteins localized specifically to cell membranes.
Alternatively, depending what you need to measure and how many measurements you need to make, it may be far more economical to manually trace.

You can try color threshold tool.. the green color can be segmented in Lab
color space by setting the suitable limits of a* channel..
