Gel analysis using ImageJ

Hi,



For Western/protein/gel analysis we do the followings:

1.       Select bands in Control and experimental lanes on the image of the
gel using Rectangle tool

2.       Then ImageJ generates Plot of individual peeks.

3.       Then we outline each peek and wand tools generates table of
Results, which sequentially numbers peeks and provides the number called
“Area”.



My questions:

I guess that “X” values, used by software to generate the Plot, are the
coordinates of the line crossing the band sequentially along the longest
side of rectangle.

Q1: Does anybody knows/explains to me *how ImageJ generates “Y” numbers on
the Plot? *

Q2: What is the meaning of those *numbers used as “Y” values on the Plot*?
