How do go about to get the grey value (or color intensity)of the red lines?

Hi,

please i am toltally new to this, my current project pushed me into attempting to use ImageJ. As you can see, I need to get the grey value of the test lines of this immunochromatographic strip. How do i go by measuring this and ensuring that the values am getting only represent that of the coloured line?

Greetings!

Before tackling your task, please make sure you use reproducible and
defined lighting and a professional camera of known gamma that is
mounted on a stable support. Image format must be either raw, TIF or
another non-lossy format.

If you don't comply to these requirements you will not be able to get
quantitative measures.

1. Light colour interacts with the colour of your specimen
    So you need to know the color of your light or
    you need to have a colour reference table.
2. The gamma of a camera should be one or known.
    If it is not one but known, you can apply gamma correction.
    If you don't care about the gamma, your measurements are
    worthless.
    (In any case don't use a cell-phone camera.)
3. Lossy compressed images such as JPG show (colour) artifacts.

Greetings,

Preface: I do not know immunochromatographic and how color translates to
grey value in this context.

If you draw a line ROI over the red strip and then Analyze>Plot
Profile(Ctrl-K) you will get a graph of the Gray Value across that line.
In the Plot window there is a List button to get the numerical values.

Image>Type>RGB Stack will convert the single color image into separate
red,green,blue component grey scale image stack.
