Ion,

after setting the proper scale, do the following:

1.
 From "Image > Lookup Tables" choose "Invert LUT".
2.
 From "Image > Adjust > Threshold..." set the threshold somewhere
between 0 and 255. (My dialog opens with a suitable choice.) Then click
"Apply".
3.
 From "Analyze > Set Measurement" check "Area" and "Limit to threshold".
4.
 From "Analyze" choose "Measure".

The value I get is:
image2.tif 4807.915

Hi Ion,

One further point. I think you need to measure the total area occupied by the tissue as well since you have a lot of white space around the section and if you are measuring multiple sections, they may be different in size, which will affect your comparisons. Area fraction may be worth considering.
