Immunohistochemistry staining

Hi everybody,

I am new on ImageJ.
I have to analyse the total area of the epithelium of some airways treated with immunohistochemistry for a protein,stained with haematoxylin,and then do a percentage of the areas stained with DAB.
I have to analyse only the staining in the epithelium trying to eliminate the one in the lumen of the airway (it is a mucin immunohistochemistry staining and it's not specific only for the epithelium).
I wanted to know if there was a specific way to do that.
