Dear Sir,
We want to measure the persistence length of a DNA molecule from an atomic
force microscope (AFM) image. For that we have to get the contour of that
molecule first and then we have to import the XY coordinate values to the
Origin software.
We can get the approximate DNA contour by segmented line tool of ImageJ
software. For that we open an AFM image as a JPEG file. Then by opening
the Set scale window we put the known distance of a line drawn on the
image and pixel aspect ratio 1.0. Then we draw a line by taking segmented
line tool and then Analyze>tools>save XY coordinate to save that file. But
the saved file don’t show the XY coordinate because firstly when import
the data to the Origin it shows that range is too big and moreover when we
take few data from that saved XY values it doesn’t give the expected
contour.
Please suggest how we can get that DNA contour to the Origin from ImageJ
software?
