Limb Bud shape tracing/comparison question

Hello everyone, I have what is most likely a simple question. I'm interested in mapping the shape change in mutant limb buds over time. This will require that I compare these limb bud's shapes with later stage mutants as well as wild types ect. I was wondering what would be the best way to do this? Simple shape tracing on the images? And if so, what would be the best way to compare them? If anyone has done this before or anything like this and could help me it would be greatly appreciated. I'm using imageJ and I also have access to an older version of photoshop. Thank you all for your help and time.

Hi Kim,
Have you already imaged these samples or are you about to begin imaging?
A key part to good image analysis is to start with good images.
if you are about to begin imaging then to be able to make comparisons
between two images ideally you need to have the spatial relationship
between object and camera the same for each image session.
A calibration scale can help to compensate for changes in
magnification but not for out of plane orientation.
An additional way to improve your imaging is to control the background
to your object by imaging against a contrasting uniform background.
This will allow you to more easily separate object from background using ImageJ.
Also use diffuse illumination, a shadow or reflection can effect the analysis.
Can you upload a typical image of what you want to image and measure
and I'm sure the community will make further recommendations

Thanks for the tips, I haven't begun imaging yet but I will soon. I'll definitely post some pictures when I have them but in the meantime thank you very much for the help!
