Macro to select ROI around specific stain

Hi Everyone,

I am working on renal tubules, and would like to quantify the number of nuclei staining positive (red fluorescence) for our protein of interest in specific nephron segments that are stained by an apical marker (green fluorescence), and there will be multiple of these in any given image. What I had thought of doing was to use the green channel to create selections of areas whose edges are defined by being a given distance from any green stain - rather like using a point to draw a circle of a certain radius, but in this case the "point" would usually be more like a line, resulting in an oval ROI/selection.  I'm attaching a picture (not sure if it will work) of what I mean, so I would want to count the red particles within something like the white selection.  I know I can create a selection by capturing all the signal that between set thresholds, then use that selection on another channel, but is there any way to automatically make that selection not just restricted to the signal, but to include an additional area around the signal?

would it help to simply enlarge an initial selection?

If yes, go to

Edit > Selection > Enlarge...

If needed, this operation can be recorded for macro programming.
