Measures of blur, goodness-of-alignment, image variation

Dear List,

I am looking for a statistical measure of image differences in a couple of
different respects:

--to quantify the goodness-of-alignment between the various images, perhaps
by z-projecting the images and then characterizing the blurriness, I guess
normalized to a single image in the series. Is this ill-advised, i.e., is
there a better way to evaluate this alignment? It might be nice also to get
this value per pixel, to be able to rank which pixels are most consistent.

--to determine the variability of the cells in the images over time, i.e.,
how well do the cells stay put? This would, I guess, also determine a
maximum value for the alignment above, as one cannot align images perfectly
unless they are identical.

I assume this trail has been blazed--can anyone help me out here?
