Dear list members:
I am to be involved into a project working on counting living and dead bacteria (colored red and green by some substance) on images obtained with a microscope. I have a lot of experience on using ImageJ on x-rays, single images, CT stacks and so on, but have never worked with particle counting on color images. I know there are some plugins but I am not sure which and how I should use them.
The images have black background and red and green ovals (bacterias), but there are some problems on counting them automatically:
- the red color contains a lot of green
- bacteria outside the sharp layer are visible (but smeared out)
- some fields with red og green shadow around bacteria groups
I have some ideas how to solve the problems. But I wonder if anybody who has experience with such work could give me advice about literature. Textbooks or papers which could give me some background of the field and methods.
