Problem with 3D Objects Counter

María Victoria Pepe
Hi, I'm having a problem with the 3D Object counter tool. I´m working with
Z-stacks of around 0,5 Gigabyte, and I've noticed that some objects present
in the original image, do not show up in the object map, even though their
fluorescence intensity is way above the threshold and the size is similar
to other objects that appear in the object map

What can Ido?

Thanks

Hi Maria,

Did you check that the objects are not touching the edges. If you
checked "exclude objects on edges" it will also remove the objects
touching the edges in Z, meaning objects is first or last slice will be
removed.

hope this helps

best,

Thomas
