RGB Profiler values

Darren David Thomson
Hi

I'm currently running an experiment that looks at membrane channel
internalisation over time.

i have a 2 channel image. RED with my membrane protein and GREEN with
GFP (which stays inside the cell at all times due to being insoluable)

I did an RGB profile of the a line across the middle of the whole of
cell of a merged G/R image and it gave me a great graph showing the
RED peaks outside the cell (at time zero) and green peaking inside the
cell.

I now need to get the number values out of this line that i have drawn
across the cell. when i click MEASURE it only gives me grey scale
values, not the separate green and red values.

My aim would be to measure the red values at the membrane compared to
inside the cell over a time course of incubation.

Thanks for you time,

regards

Convert the RGB image to composite color using Image>Color>Make Composite and the Measure and Plot Profile commands will give separate red and green values. Enable "Display Labels" in Analyze>Set Measurements and, when using the Measure command, the channel ("Red" or "Green") will be shown in the Results table "Label" column.
