As other respondents have noted it is not always simple.
However, in my experience the Stack Focuser plug-in works very well.

Caveats.

If your lens has distortion (barrel or pincushion) then the outer parts of
an image will not be portrayed well. Distortion will result in the
production of concentric fringes around regions of strong contrasts.

If the images are not precisely one on top of the other (i.e. no wobble in
Z) the results will be off.

If the exposure varies between images the results may be not so good.

If there are areas of overexposure (saturated white) or underexposure
(saturated black).  A series of contoured fringes will be produced around
each of them.

Also if the object is composed of smooth detail free (no contrast) surfaces,
such as a crystalline surface, or a set of perfect tilted mirrors, then the
task of finding the in-focus pixels becomes challenging.

And as Gabriel Landini comments, if the subject is translucent or
transparent your mileage may vary.
