Set Scale problem

I'm new to ImageJ but only need it to measure distances in images. I did
the tutorial and figured it out and it was going fine. Then in the Set
Scale box I accidentally clicked on "Click to remove scale" button.

 

Now when I draw my line of known distance and go to Set Scale to fill in
the known distance and unit of measure it does not show me how many
pixels are equal to my known distance and just above the OK button it
says, "Scale:<no scale>"

 

How do I get back to the normal setting?

Hello,

I'm writing to see if anyone can tell me whether or not Image J is "ICC
aware."

I'm also interested in whether or not ImageJ uses the monitor ICC
profile for displaying images, or whether it uses some other means for
displaying.

For example, if I have calibrated a monitor so that whites are neutral
(an equal amount of red, green and blue) in color images and have
calibrated it so that shades of black just above zero (0) can be
distinguished, will the calibration (as an ICC profile) be used for
displaying images in Image J, at least with 8-bit (gray) or 24-bit
(color images)?  I get it that 12- and 16-bit images are auto-scaled for
display in Image J.

I'm in the Windows environment.  I also get it that the newer Mac OS
color manages across all software programs, so speaking about Macs may
be another subject altogether.

Any insights would be very helpful!

If you have not explicitly saved the image file, the scaling info will
still be available - all you need do is use the revert option from the
file menu, or reload the image from the original file. If you have saved
over the original file, you will need to calibrate the image possibly
using Analyze - Set Scale.

Please clarify what you mean by "no pixels shown in set scale window". I
assumed that your images come calibrated from your imaging system
(microscope?). If this is true, opening the original image file should
have shown, above the image, in addition to the pixel size of the image,
additional info such as 128 nm x 128 nm,indicating the spatial extent of
the image. If you see this, it implies that the scaling information is
included and you can make measurements as before. If you do not see this
information, you need a way of putting in scale information, which is
what I referred to as done by using the Set Scale option. Before using
the Set scale, you need to make a line selection of a known distance on
the image and enter that value and unit in the set scale dialogue box.

Please copy your response to the list so that others who may have better
or more appropriate advise can also respond.

The Set Scale box has as the first line of information "Distance in Pixels." That box has a positive value when I had selected a known distance. Then I would type in the known distance and units. After that line segments could be measured based on that set scale.

What has happened is that the "Distance in pixels" box never fills in when I mark out a line segment of known length. Therefore, I am unable to get a true measurement of anything. Also, just above the "OK" button at the bottom it reads "Scale: <no scale>" even after I try the line segment of known length.

I hope that clarifies my problem.
