area measurment

area measurment

please i need help how to measure area

i followed these steps:
1:split channels
2: worked on the green one, then i made subtract for the background
3: from image i choose adjust then threshold
4: then from
5: edit, selection , create selection
6: analyse, set measurement  ( checked limit to threshold and area and
directed to the green photo)

7: analyse, measure where the problem is,,,  the read i got is the whole
image area
and when i use anaylse particles instead it doesn't give me a correct
result at all as sometimes i got the area as 100 %

please help me

Hello Thuraya, I hope this help you:
          first I would set the scale of the image (Analyze; Set
Scale). Then do your steps and measure your selection. The value of
%Area represent the percent of the area occupated by pixels with 255.
If you multiply the value of the Area (total selection area) by %Area
(which is the percent of that area with a value of 255) divide by 100
you get the area of the signal.
