I am a new user of Image j and I'm having trouble with the colour deconvolution plugin.
I have single stained 40X images of DAB and hematoxylin of which I processed to subtract the background so that empty areas appear white. I pasted selected areas from both single stained images into a single test image (new RGB image, 2 slices), I then follow the instructions for the plugin by selecting the ROIs. But the separation is never optimal, I have repeated it several times selecting different ROIs, but I never get a clean separation.
Maybe I am missing some information/steps, hope someone can help advise me,
