corelation between pixel value to real scale

Hi everybody,

I've 8 bit SEM image. I calculated surface roughness using plugin. But it gives Rq value in pixel. How can I corelate this to real scale ( micrometer) ?
Please give some advice.

your name Charles Lessman
With image open, select set scale from analyze menu and fill in drop
down. Then redo your calculations.

I assume that if you have 8-bit SEM images then you are just calculating the greylevel variation of the images, not the real roughness values. For calculating the roughness (e.g. Rq) you need to reconstruct the surface, using e.g. SEM stereo imaging.

The Analyze/Set scale... will only calibrate the x,y dimensions, not the heights (z).

Could you tell me please what is the pixel aspect ratio? Is it the ratio of two values shown by cursor when I move it on picture?
