measuring distances between lines

I want to segment images/stacks with segmented lines and measure the
distance between lines continuously to get
1. Thickness measurements of layers within single images
2. Volume measurements of these layers within stacks
Two questions:
1. How can I perform such distance measures within ImageJ?
2. Can I apply these to a stack for volumes?
