ratiometric images

shilo
Dear list
I'm sending the massage again because I wasn't subscribed.

I would like to produce ratiometric images from two stacks. The two pictures come from confocal microscope and each one of them is from different excitation wavelength.
I was trying to make it with the calculator plus tool (divide), but I got image with 0,1,2 values and not continues values.
In the next step I would like to have colors that represent the ratio values and scaling bar

Thanks for your help

Re: ratiometric images

Hi Shilo,

use Process>Image Calculator and check "Create New Window" and "32-
bit Result".
8-bit or 16-bit images can hold only integer numbers; "32-bit" is the
floating point format.

For your other questions, see Image>Lookup Tables and
Analyze>Tools>Calibration Bar (grayscale calibration) and
Analyze>Tools>Scale Bar (spatial calibration).

Hello Shilo,

I'm guessing that if you first converted the images to floating point (menu: Image->Type->32 bit), you would get closer to the results you expect.
