where to begin..

Hello,

My name is Richard, I am a masters student working on a project which involves TLC quantitation using ImageJ. After exploring ImageJ and all its functions, I'm finding myself VERY lost. I need help, so i have come to this forum in hopes to gain some knowledge and direction in using the program to my advantage.

Currently, I have been taking photographs of developed TLC plates as a RAW image and converting them to .tif files. I then upload the files to Image J where I attempt to convert the image to grayscale. From there, I am lost... Essentially, what I am attempting to do is create a plot of the spot (substance on TLC plate) on the plate. In doing so, I am trying to either find the area under the curve, or optical density (haven't decided which would be the best course of action), but either way I cant seem to figure out what to do. I know i need to calibrate as well as set a background? From there, my goal is to build a calibration curve and be able to determine an unknown concentration from the standards that i have made (with the values obtained from ImageJ). Im stuck, so much so, that I'm losing sight of what I'm really trying to do here. If anyone has any idea what I may be trying to achieve and could help me, in any way, I would really appreciate it...at the very least, put me in the right direction.
