Segmentation of phase contrast images

Dear All,

 

I'm trying to segment phase contrast images of adherent cells but I'm
having some trouble doing it. At the moment, I don't need to know how
many cells there are but simply the area covered by the cells.

 

I would appreciate any suggestions.

You might try edge detection followed by filling holes.

If the question is so simple why do you need segmentation.
You can get by generating a Grid of points and then simply counting
how many crosses hit any of the cells.
In such way you ge the area estimate of the cells. then simply subtract
the area which is obviously void of cells (I mean
large empty fields) and take the ratio and voilà -
you ll have the area fraction.
I did the same while counting muscle cells
in cross sections (quite difficult to segment).
