Trichrome Stain Analysis

Dayana Webster
Hello,
After staining with Masson's Trichrome stain, I am analyzing
immunohistochemistry images for the amount of blue collagen vs. red muscle
fibers.  I am having trouble finding a plug-in the can help me distinguish
and count the blue vs. red areas in the images.  Could anyone help me?

Thanks,

Dayana,

Try G. Landini's *Threshold_Colour*  plugin. You can get to it via
ImageJ/Plugins. Then look up Landini's page. The plugin will compute HSV
histogrms from an RGB image. I found that varying the threshold on the Hue
histogram separates out color regions in the image nicely. This should be a
good start point.

Trichrome segmentation is an almost archetypal application of k-means
clustering. There is an ImageJ plugin that implements this at:

http://ij-plugins.sourceforge.net/plugins/clustering/index.html

It is used as an example in this paper:

http://www.biotechniques.com/biotechniques/BiotechniquesJournal/supplements/2007/July/ImageJ-for-microscopy/biotechniques-42894.html?pageNum=2

Hope that helps!

Francis
