basic question

Hi there.
This is actually quite a basic question.

To measure fluorescent intensity, which of the measurement is used for analysis? Is it the 'integrated density' or the 'mean gray value'? Or is it something else?
I'm confused. Please help.

You can take whatever you want. But the integrated densityis  area-dependent, this is, why i always take the mean grey value.
