Import your time series of images as a sequence stack into ImageJ
(File->Import image sequence).
Draw a region of interest box on the desired location of your image
on one of the images in the stack.
To plot the average pixel intensity in in the region of interest as a
function of time, choose the Image->Stacks->Plot z-axis profile
function.
The graph can be exported as a text file for Excel spread sheets if
desired.
The same operation can be performed on a single pixel if you choose
the point-picker tool instead of the region of interest drawing tool.
